Background:Lymphotoxin-? (LT-?), also known as TNF–?, is a member of the TNF superfamily of proteins. NK cells, T cells, B cells, and lymphoid tissue-inducer cells express LT-?. LT-? can be secreted as a soluble homotrimer or form membrane bound heterotrimers with lymphotoxin-? (LT ?1?2 or LT ?2?1) which can be cleaved from the cell surface by matrix metalloproteases. Soluble LT-? binds to and signals through TNFR1/TNFR2, activating the canonical NF-?B pathway. In contrast, LT ?1?2 heterodimers bind to the LT?R receptor and activate the noncanonical NF-?B pathway. As a result, LT-? and TNF-? have overlapping functions. Soluble LT-? and LT ?1?2 play key roles in lymphangiogenesis. The LT ?1?2/LT?R axis is essential for the development of lymphoid tissue.
