The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

Assay procedure summary: 1. Prepare all reagents, samples and standards;
2. Add 100µL standard or sample to each well. Incubate 1 hours at 37°C;
3. Aspirate and add 100µL prepared Detection Reagent A. Incubate 1 hour at 37°C;
4. Aspirate and wash 3 times;
5. Add 100µL prepared Detection Reagent B. Incubate 30 minutes at 37°C;
6. Aspirate and wash 5 times;
7. Add 90µL Substrate Solution. Incubate 10-20 minutes at 37°C;
8. Add 50µL Stop Solution. Read at 450nm immediately.

Test principle: The microplate provided in this kit has been pre-coated with an antibody specific to thrombin. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to ATIII. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain TAT, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of TAT in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Research Area: Signal transduction;Enzyme & Kinase;Metabolic pathway;Hematology;

References Citing This Product: Genetic repression of mouse VEGF expression regulates coagulation cascade.

Treatment with unfractionated heparin attenuates coagulation and inflammation in endotoxemic mice

Tissue factor and PAR1 promote microbiota-induced intestinal vascular remodelling

Host Defense Peptides of Thrombin Modulate Inflammation and Coagulation in Endotoxin-Mediated Shock and Pseudomonas aeruginosa Sepsis

Novel evidence that crosstalk between the complement, coagulation and fibrinolysis proteolytic cascades is involved in mobilization of hematopoietic stem/progenitor cells (HSPCs)

Thrombomodulin Improved Liver Injury, Coagulopathy, and Mortality in an Experimental Heatstroke Model in Mice

Evidence of a Pivotal Role for the Distal Part of the Complement Cascade in the Diurnal Release of Hematopoietic Stem Cells Into Peripheral Blood

(S)YS-51, a novel isoquinoline alkaloid, attenuates obesity-associated non-alcoholic fatty liver disease in mice by suppressing lipogenesis, inflammation and coaCavia (Guinea pig )lation

Coagulation factor XI vaccination: an alternative strategy to prevent thrombosis.

Platelet-localized FXI promotes a vascular coagulation-inflammatory circuit in arterial hypertension

Transduced Tat-aldose Reductase Protects Hippocampal Neuronal Cells against Oxidative Stress-induced Damage

The impact of dabigatran treatment on sinusoidal protection against hepatic ischemia‐reperfusion injury in mice

Doxorubicin contributes to thrombus formation and vascular injury via enhancing platelet functions in vivo

Complement factor D is linked to platelet activation in human and rodent sepsis

//cdn.shopify.com/s/files/1/0558/8588/9636/files/no_image_default_image-jpeg_d6e18437-54dd-4c40-a9f0-e66c13d9504d.jpg?v=1783135777
