An antibody specific for MAP4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMAP4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MAP4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAP4 bound in the initial step. This protein contains a domain similar to the microtubule-binding domains of neuronal microtubule-associated protein (MAP2) and microtubule-associated protein tau (MAPT\/TAU). This protein promotes microtubule assembly, and has been shown to counteract destabilization of interphase microtubule catastrophe promotion. Cyclin B was found to interact with this protein, which targets cell division cycle 2 (CDC2) kinase to microtubules.The phosphorylation of this protein affects microtubule properties and cell cycle progression.
