After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of KEAP1 bound in the initial step. Two alternatively spliced transcript variants encoding the same isoform have been found for this gene.Kelch motifs of Keap1 interact with the Neh2 domain of Nrf2 and that this interaction is required for the repression of Nrf2 by Keap1.
