An antibody specific for PRDM9 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPRDM9 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PRDM9 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PRDM9 bound in the initial step. PR domain-containing proteins, such as PRDM9, are often involved in transcriptional regulation.\nThe deduced 862-amino acid Meisetz protein contains a PR\/SET domain in its N-terminal portion and a zinc finger domain containing multiple C2H2-type zinc fingers in its C-terminal portion. Hayashi et al. (2005) also cloned 2 Meisetz splice variants that encode shorter isoforms lacking the zinc finger domain. The PR\/SET domain of Meisetz is highly conserved in human PRDM9.
