An antibody specific for VCAM1\/CD106 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyVCAM1\/CD106 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for VCAM1\/CD106 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of VCAM1\/CD106 bound in the initial step. The rat and mouse SVCAM-1 proteins share approximately 76% amino acid similarity.\nVCAM-1 is a member of the Ig superfamily and encodes a cell surface sialoglycoprotein expressed by cytokine-activated endothelium. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
