An antibody specific for MAPKAPK5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMAPKAPK5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MAPKAPK5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAPKAPK5 bound in the initial step. In response to cellular stress and proinflammatory cytokines, this kinase is activated through its phosphorylation by MAP kinases including MAPK1\/ERK, MAPK14\/p38-alpha, and MAPK11\/p38-beta. In vitro, this kinase phosphorylates heat shock protein HSP27 at its physiologically relevant sites. Two alternately spliced transcript variants of this gene encoding distinct isoforms have been reported.MAPKAPK5 encodes a predicted 471-amino acid protein that shares 20 to 30% sequence identity with RSK, MNK, and MAPKAPK kinases.
