After removing any unbound substances, a biotin-conjugated antibody specific for KRT20 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of KRT20 bound in the initial step. Results strongly suggest that transcriptional regulation of keratin genes in the intestinal epithelium occurs at the level of both immature and terminally differentiated epithelial cells, and is tightly regulated during both fetal development and crypt-to-villus differentiation of the intestinal epithelium.\nCK20 has recently been reported to be useful to distinguish between primary and metastatic lung adenocarcinoma.
