After removing any unbound substances, a biotin-conjugated antibody specific for DKK1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DKK1 bound in the initial step. Elevated levels of DKK1 in bone marrow plasma and peripheral blood is associated with the presence of osteolytic bone lesions in patients with multiple myeloma. Sequence analysis predicted that the 266-amino acid DKK1 protein contains a 31-amino acid N-terminal signal peptide, 2 clusters of cysteine residues, and a potential C-terminal N-glycosylation site.
