After removing any unbound substances, a biotin-conjugated antibody specific for MAPRE1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAPRE1 bound in the initial step. It is involved in a variety of cellular processes, including establishment and maintenance of cell polarity, search and capture of chromosomes during mitosis, and positioning of the mitotic spindle during asymmetric cell division.\nThis microtubule localization was abolished in the presence of the microtubule-destabilizing drug nocodazole; upon drug removal, the microtubule distribution of EB1 was recovered and EB1 fluorescence concentrated at the microtubule-organizing center.
