The Yeast Genomic DNA Extraction Kit provides a simple and rapid method for high quality genomic DNA purification from yeast. The kit procedure uses the silica-gel-membrane spin column technology for simple and fast isolation of Genomic DNA without phenol/chloroform. Homogenization is not necessary since cells are directly lysed by Proteinase K and lyticase.
Lyticase (10 U/ul), 3000 U or 10,000 U Pack Sizes Lyticase from Arthrobacter luteus is used to lyse yeast cells' difficult-to-disrupt cell walls. DNA can be extracted from yeast by using lyticase or equivalent enzymes to induce partial spheroplast formation; spheroplasts are subsequently disrupted to release DNA.
